The tautomerase superfamily (TSF) consists of more than 10,000 nonredundant sequences found throughout the biosphere. Typified by the initial enzyme characterized, 4-oxalocrotonate tautomerase (4-OT), the superfamily members are composed of various fusions and oligomers of a small structural subunit and typically require a catalytic N-terminal Pro for function. Based on sequence similarity, superfamily members fall into five major subgroups with different chemical capabilities and unique structure-function relationships. The few characterized members catalyze a diverse range of chemical reactions, but the full scale of their chemical capabilities and biological functions remains unknown.
