The tautomerase superfamily (TSF) consists of more than 10,000 nonredundant sequences found throughout the biosphere. Typified by the initial enzyme characterized, 4-oxalocrotonate tautomerase (4-OT), the superfamily members are composed of various fusions and oligomers of a small structural subunit and typically require a catalytic N-terminal Pro for function. Based on sequence similarity, superfamily members fall into five major subgroups with different chemical capabilities and unique structure-function relationships. The few characterized members catalyze a diverse range of chemical reactions, but the full scale of their chemical capabilities and biological functions remains unknown. The Download Archived Data tab provides sequence similarity networks for the superfamily and its subgroups, along with annotation data and fasta sequences of the set. Similarity networks are also provided for figures 2, 3a, 4b, and 3, colored and highlighted as they are shown in the classification paper, “A global view of structure-function relationships in the tautomerase superfamily” (PMID 29184004).
